吖啶橙荧光法测定牛血清白蛋白

摘    要:吖啶橙在十二烷基硫酸钠介质中存在单体、二聚体形态之间的平衡,蛋白质的加入可使吖啶橙的平衡向单体转化,导致体系荧光增强,其荧光增强的强度△I与蛋白质的浓度在一定范围内呈线性关系,可用于蛋白质的定量测定。对于牛血清白蛋白,线性范围为0-34μg·mL^-1,检出限为0.126μg·mL^-1。建立了荧光光度法测定痕量牛血清白蛋白的新方法。

Abstract:Acridine Orange has an equilibrium of aggregate forms in the presance of sodium dodecyl sulfate, but the equilibrium can shift when the protein is added to the system, thus leading to the enhancement of fluorescence intensity. The change of fluorescence intensity shows a good liner relation with the concentration of the protein in a certain range, which can be applied to the determination of trace amount of the protein. For the Bovine Serum Albumin, the linear range of the determination is 0---34 μg.mL^-1, and the detection limit is 0.126μg.mL^-1. A new method of fluoremetde determination of Bovine Serum Albumin was developed.
