摘    要:以苹果渣为原料,用乙醇溶液提取根皮苷,以高效液相色谱法测定根皮苷含量.利用BoxBehnken中心组合设计原理,应用响应面实验优化方法确定最优提取条件,同时应用自由基清除实验考察根皮苷提取物抗氧化性.实验结果显示,最优提取条件为乙醇浓度60%,料液比1∶10,提取温度70℃,提取时间5h.在此条件下根皮苷的提取量可达到2.79mg/kg,各因素对吸附率影响顺序为:提取温度>料液比>提取时间>乙醇浓度;根皮苷粗提物的抗氧化性明显高于苹果渣,表现了较好的抗氧化能力.

Abstract:Abstact:The apple pomace was used as raw material,and the phlorizin was extracted from it by ethanol solution and was tested by HPLC.The optimization condition of extraction condition was confirmed using response surface methodology with Box-Behnken design,and the antioxidation of the phlorizin extract was assayed by free radical scavenging test.The result shows that the optimized extraction condition is:the content of ethanol solution,the ratio of pomace to ethanol solution,the temperature and the time are 60 %,1:10,70C and 5h,respectively,in this condition the extracted phlorizin quantity could reach to 2.79mg/kg,and the sequence of each factor effecting on extracted phlorizin quantity is:temperature>the ratio of pomace to ethanol solution> time>the content of ethanol solution.The phlorizin extract shows more obvious antioxidaton than apple pomace,exhibits good antioxidation capacity.
