摘    要:巨噬细胞是机体重要的免疫细胞,具有免疫调节、抗感染和抗肿瘤等重要功能。快速、有效地分离得到高纯度的人外周血单核细胞,是研究巨噬细胞功能的前提。本文利用细胞大小及胞内颗粒度属性对单核细胞与其他白细胞组分进行区分,通过流式细胞分选,获得了较高纯度单核细胞。单核细胞表面标志物CD14染色分析显示,该方法对单核细胞的分选效率达85%以上。进一步实验证明,分选得到的单核细胞能够在体外正常分化为巨噬细胞。

Abstract:Monocye-Macrophages are key players in the immune system and dysregulation of its function will lead to uncontrollable infections, tumor and autoimmune diseases. Isolating purified monocytes from human peripheral blood is the key step for macrophages related study. Here we show a method that we used to isolate cells with the flow cell sorter by the size and granularity of cells. Sorted cells were then confirmed by staining of CD14, the surface marker of monocytes and the purity of the cells is more than 85%. This simple and efficient method can be used to isolate pure monocytes for functional research.
