天门冬多糖的分离纯化及其部分理化性质

摘    要:中药天门冬冷水提取物,通过脱蛋白、丙酮分级及柱层析纯化,得到三种级份多糖:PA-W、PA-I和PA-Ⅱ,凝胶柱层析测定,它们的分子量分别是158万、219万和219万。三种多糖中均含有葡萄糖,甘露糖和半乳糖,但各自的摩尔比相差很大。此外,PA-Ⅱ中还含有一种未知的糖醛酸。高碘酸氧化、Smith降解、红外光谱和13C核磁共振谱表明,PA-Ⅱ含α和β两种构型,具有1→4和1→6两种糖苷键,二者比例为5~6:1,是一种具有高度分枝结构的半乳甘露聚糖。

Abstract:The rough polysaccharide was extracted with cold water from tradition Chinese medicineAsparagus Cochinchinensis,and three polysaccharides,PA-W, PA-I and PA-Ⅱ with Mws 1.58×106, 2. 19×106and 2. 19×106respectively,were isolated by deproteination, acetone fractionation and column chromatography. They all contain glucose, mannose and galactosee, but the rstios are different. In addition, PA-Ⅱcontains a kind of unidentified glycouronic acid,Periodate oxidation, Smith degradation and spectra of IR and 13C NMR reveal PA-Ⅱ owns αand β configurations, and has 1→4 and 1→6 linkages in the ratio of 5~6: 1.
