硫酸化茯苓多糖药理性质研究

摘    要:探讨硫酸化茯苓多糖(SP)的急性毒性强弱、药代曲线及其对肾功能的影响.通过灌服不同剂量SP,计算SP的半数致死量(LD50)等;用分光光度法测定不同时间段家兔血浆中SP浓度;腺嘌呤复制CRF模型,观察SP对CRF大鼠肾功能的影响.给药后在测定的时间段内(0.33~8 h)SP在血浆中均有分布,1.5 h时达最大;用药30 d后,SP组的尿量、尿肌酐排出显著增加,尿蛋白排出则明显减少,血清Scr和BUN水平有明显的降低,TP和ALB明显增加.因此,SP属低毒类物质;药代出峰时间合适;对大鼠CRF有明显防治作用.

Abstract:Toxic strong and the weak and curve of medicine metabolism and influence to kidney function of sulfation pachymaran was discussed. Worked out the LD_(50) by ig; determined on the determination of sulfation pachymaran in plasma of the rabbit by spectro-photometric method; Investigated the effect of sulfated pachymaran(SP) in rats with chronic renal failure(CRF) induced by adenine. The LD_(50) of the sulfation pachymaran was 7 .358 g·kg~(-1), and 95% of LD_(50) within the average of(7.358±0.894)g·kg~(-1) , after the sulfation pachymaran was given to the rabbit by ig, in the range of 0.33-8 hours, the sulfation pachymaran was found in the in plasma of the rabbit and the concentration reached the peak at 1.5 h., after 30 days treatment, SP significantly increased the volume of urine and the contents of urinary creatinine, serum TP and ALB, while declined the contents of urinary protein, serum Scr and BUN(P<0.01). The level of serum TP was nearly equivalent to that of normal control group. Also, SP alleviated the injury of the kidneys. Whose toxicity was very low, Medicine metabolism appears the peak to be appropriate, SP appeared to have a preventive and therapeutic activity against chronic renal failure.
