左手螺旋Z-DNA的研究:人工合成的Oligod(G—C)_n在PAGE中的异常泳动行为

摘    要:为研究DNA的B,Z构象的相互转换,我们人工合成了dG-dC交替互补顺序的六聚体与十二聚体,在用PAGE法纯化过程中,发现5',3'-末端为双羟基的寡聚体的泳动速率是d(G-C)_3>d(G-C)_2>d(G-C),5'—末端带磷酸基的相应寡聚体的泳动速率则是:d~*(G-C)>d~*(G-C)_2>d~*(G-C)_3,二者行为正好相反,而5',3'—末端为双羟基的d(G-C)_5与d(G-C)_6和5'—末端带磷酸基的十聚与十二聚体在PAGE中的泳动行为是一致的。由此推测5',3'—末端为双羟基的寡聚体在PAGE中的异常行为的转折点在六聚到十聚之间。

Abstract:All oligodeoxynucletides were synthesized by Solid-Phase-Phosphite-Triester method, and purified with PAGE. We observed that the electrophoretic mobility was oligod (G-C)_3>d (G-C)_2>d (G-C), just in reverse order with regard to oligomtrs containing terminal phosphate resides. The electrophoritjc behaviour of oligod (G-C)_6,d (G-C)_5carryjng the hydroxy or phosPhated groups at terminal was the same. Hance, we suggest that the turning point was probably locating between 6th mer to 10th mer.
