人组氨酰-tRNA合成酶基因的克隆与表达

摘    要:用RT_PCR方法从人胎盘总RNA中获得编码人组氨酰_tRNA合成酶(Jo_1)基因的全序列,选用IMPACT_CN系统中的pTYB11,构建克隆与表达载体,转化大肠杆菌ER2566.经筛选与鉴定,得到3个阳性重组子,对其进行诱导表达获得Jo_1融合蛋白,western_blotting检验结果显示,该融合蛋白具有Jo_1免疫原性和免疫特异性.

Abstract:Using RT-PCR technology the full length DNA sequence coding for human histidyl-tRNA synthetase was obtained. The pTYB11 was used in construction of expressing vector. The recombinant plasmid constructed was transformed into ER2566 and the system was used to express fusion Jo-1 antigen. At last, the author got 3 recombinants that could successfully express the fusion protein. Meanwhile, the expressed fusion protein could be identified by western-blotting using anti-Jo-1 antibody.
