玉米Mn-SOD的分离纯化与性质分析

摘    要:利用金属螯合亲和层析,结合DEAE-纤维素离子交换层析,从玉米匀浆液中分离纯化到电泳纯的Mn-SOD组分.双向电泳和SDS-PAGE分析表明:玉米Mn-SOD的亚基分子质量为26.2 kD,等电点为5.3;热稳定性、酸碱稳定性高于玉米Cu/Zn-SOD.

Abstract:Metal chelate affinity chromatography and DEAE-cellulose ion-exchange chromatography were used to separate and purify SOD from maize in this research.A electrophoretic pure Mn-SOD component was obtained.The SDS-PAGE and 2-D electrophoresis showed that the subunit of the Mn-SOD was 26.2 kD and its isoelectric point was 5.3.The experimentation of stability showed that the Mn-SOD component had higher thermal stability and pH stability than Cu/Zn-SOD.
