胀果甘草愈伤组织生长和多糖合成的调控

摘    要:为了研究培养基基本成分、碳源以及离子浓度对甘草愈伤组织生长和多糖合成的影响,以MS,6,7-V为基本培养基,改变碳源、PO43-及Ca2+浓度,分析胀果甘草愈伤组织生长及多糖产量.结果表明:MS培养基附加蔗糖最利于甘草愈伤组织生长,6,7-V培养基虽利于甘草多糖的合成但不利于愈伤组织生长从而影响多糖产量.当培养基中PO43-浓度为1.25mmol/L、Ca2+浓度为2.99mmol/L时,甘草愈伤组织生长量积累最高,同时对多糖的生物合成也最为有利.高浓度的PO43-和Ca2+有利于甘草多糖的合成,但不利于愈伤组织的生长.研究结果为利用生物工程手段进行甘草多糖的产业化生产提供参考数据.

Abstract:In order to study basic composition and ion concentration of culture medium on the growth of Glycyrrhiza inflata callus and effects of polysaccharide synthesis process,MS and 6,7-V were used as basal medium to study their regulation on growth and glycyrrhiza polysaccharide biosynthesis of Glycyrrhiza inflata callus,and the effects of the types of carbon source and PO43-and Ca2+ of medium were investigated.The results showed that among the tested basal medium,MS with the addition of sucrose was the best for callus growth,6,7-V was most suitable for glycyrrhiza polysaccharide production.The optimal concentration of PO43-and Ca2+ of medium for biomass accumulation of callus and biosynthesis of glycyrrhiza polysaccharide was 1.25 mmol/L and 2.99 mmol/L.The higher concentration of phosphate and Ca2+ were benefit of the production of glycyrrhiza polysaccharide,but not the growth of callus.The results offered the reference data of industrialized production of glycyrrhiza polysaccharide by biological engineering means.
