摘    要:本文探讨了在大鼠背根神经节(DRG)细胞上记录离子单通道电流的方法,研究了膜片钳实验的各个环节,研究了提高封接电阻的方法。研究结果表明,单通道记录的背景噪声取决于电极与细胞的封接水平。文中还讨论了实验系统的组装,电极制备和细胞分离等技术。

Abstract:Patch clamping is a new cell electrophysiological method for single ionic channel recording from cell membrane. The dorsal root ganglion (DRG) cells are the first stage neuron of the sense system. Single-channel recording from DRG cells will be helpful to the study of the mechanism of sense information transmission and modulation.In this paper, the techniques of single-channel recording from DRG cells of adult rats including the experimental system assembly, the preparation of patch electrodes and DRG as well as the experimental procedures are studied. The formula for calculating the electrode capacitance is also given.
