荧光光谱法分析环境因素对溶藻细菌S7溶藻作用的影响

摘    要:通过对水华鱼腥藻活体进行荧光光谱分析,确定了其藻蓝蛋白特征荧光波长,建立了水华鱼腥藻生物量与藻蓝蛋白特征荧光强度之间的线性关系;同时,在改变蓝藻生长的条件下,研究不同环境因素(温度、光照条件和pH值)对溶藻细菌S7溶藻作用的影响.结果表明,采用荧光光谱法检测的藻蓝蛋白荧光强度可以反映水华鱼腥藻的生物量,其操作简单且具有较高的可行性和准确性.环境因素对溶藻细菌S7的溶藻效果影响较大,在不同环境因素下,其溶藻效果由强到弱依次为:不同温度时,30 °C>35 °C>40 °C>25 °C;不同光照条件下,黑暗>光循环>全光照;不同pH值时,pH 5>pH 10>pH 6>pH 9>pH 8>pH 7.

Abstract:Anabaena flos-aquae were scanned in vivo by fluorescence spectrum scanning to acquire characteristic fluorescence wavelength of phycocyanin and build a linear relationship between Anabaena flos aquae biomass and the phycocyanin characteristic fluorescence intensity. With the change of cyanobacteria growth conditions, the lytic effects of varying environmental factors (temperature, illumination and pH) to the algae lysing bacterial strain S7 were studied. The results show that utilizing fluorescence spectrum for detecting the phycocyanin fluorescence intensity and reflecting anabaena flos aquae biomass are simple, fast, accurate and feasible. Environmental factors affect the lytic effect of algae lysing bacterial strain S7. Levels of lytic effects are 30 °C>35 °C>40 °C>25 °C, dark>light cycle>full light, pH 5 >pH 10>pH 6>pH 9>pH 8>pH 7. For utilizing algicidal bacteria to control water bloom, exploring optimal environmental conditions for restraining the growth of those bacteria is of practical significance.
