烟草NC89与香料烟Basma种间原生质体融合研究

摘    要:NC89与Bsama的叶肉原生质体经聚乙二醇融合剂处理后,培养在改良的NT培养基及KM8P培养基上,得到了二百多块愈伤组织,分化出三种类型的苗,有两种类似亲本,而另一种不同于双亲.对此类型进行形态学观察和过氧化物同工酶检测表明,它们具有双亲的特征,并对融合过程中的一些条件和方法进行了研究和探索.

Abstract:Mesophyll protoplasts from NC 89 and Basma, which were treated with PEG, the higher concentration of Ca 2 and pH, are cultured in media of modified NT and KM8P. More than 200 callus are obtained. The fusion frequency was 15%. The callus differentiated three kinds of plantlets through culture, two of which were similiar to their parents. Morphology observation and the peroxidase isoenzyme analysis showed that they had the characterictics of their parents. Some conditions and methods were also studied in the fusion experiment.
