摘    要:目的:建立一种省时、省力的人载脂蛋白E基因多态性的检测方法。方法:采用聚合酶链—限制性片段长度多态性(PCR-RFLP)方法,先扩增出apoE基因第4外显子内的272bp片段,继以限制性内切酶CfoⅠ酶解消化,8%聚丙烯酰胺凝胶电泳,最后银染显带判定基因型。结果:电泳、染色结果清晰、特异、分辨率高。结论:该方法快速、准确、适于临床实验室中广泛开展。

Abstract:Objective: To establish a simple, accurate and practical method for detecting apolipoprotein E (apoE) genotyping. Methods: The polymerase chain re action-restriction fragment length polymorphism(PCR-RELP) was used to amply 227 base - pair DNA fragment in the fourth exon of apoE gene and to digest them with restriction enzyme Cfo I, electrophore on 8% polyacrylamaid gel for the cleavaged products, subsequently genotype was determined by silver staining.Result: The results of eletrophore and staining was clear, specific and sensitive. Conclusion:PCR - RFLP is considered a rapid and exact method,and is suit able for routine laboratories.
