高温大曲中筛选产纤维素酶的耐高温芽孢杆菌

摘    要:从高温大曲中筛选出产纤维素酶的芽孢杆菌1株,进行形态特征、生化鉴定和16S rDNA序列分子鉴定,该菌株鉴定为Bacillus cereusFrankland.对该菌株进行产酶摇瓶液体发酵,测定其内切纤维素酶活力和外切纤维素酶活力分别为260.32,87.22u.mL-1,对该菌株进行产酶固态发酵,测定其内切纤维素酶活力和外切纤维素酶活力分别为556.64,121.47u.mL-1.酶的稳定性实验显示该菌株产纤维素酶在60℃以下和在pH4~8范围内具有良好的热稳定性和pH稳定性,利用响应面法优化固态发酵产酶培养基,优化后测定纤维素酶活提高至1 643.27u.mL-1.

Abstract:The Bacillus that can produce cellulase is isolated and purified from the high temperature Daqu,based on the analysis of 16SrDNA and some biochemical and physiological characteristics,this strain was identified as Bacillus cereus Frankland.The shake flask liquid fermentation was taken to determine the endo-cellulase activity(260.32 u·mL-1),and exo-cellulase activity(87.22 u·mL-1),the solid state fermentation was taken to determine the endo-cellulase activity(556.64 u·mL-1)and exo-cellulase activity(121.47 u·mL-1).High heat and pH stability of cellulase activity are observed at temperature lower than 60 ℃ and pH4~8.After the response surface method optimization,the cellulase activity of solid state fermentation increased to 1 643.27 u·mL-1.
