单宁微球固定化酶的应用性能

摘    要:以单宁微球为载体制备固定α-淀粉酶.探讨了温度、pH值、淀粉初始质量浓度、淀粉种类等对单宁微球固定化α-淀粉酶催化淀粉水解性能的影响.结果表明,固定α-淀粉酶在25,60,90℃催化水解淀粉17h后,淀粉的水解百分率均达到95%以上,说明温度对固定α-淀粉酶的催化性能影响不大;pH对固定α-淀粉酶的催化性能影响大,最佳pH值为8;固定α-淀粉酶对可溶性淀粉、番薯淀粉、芭蕉芋淀粉的催化水解率均大于96%;固定α-淀粉酶重复5次后仍具有较好的催化效果.

Abstract:α-amylase was immobilized on tannin microspheres.The effects of temperature,pH and initial concentration of starch on the catalytic performance of immobilized enzyme were investigated.The results showed that each of the hydrolysis degree of starch hydrolyzed by immobilized α-amylase under 25,60 and 90 ℃ for 17 h was more than 95%.The effect of temperature on catalytic performance of the immobilized α-amylase was limited.The optimum of pH was 8.The hydrolysis degree of soluble starch,sweet potato starch and canna edulis starch,which was hydrolyzed by immobilized α-amylase under 60 ℃ for 17 h,was more than 96%.Moreover,the immobilization enzyme still had a good catalytic performance on hydrolysis of starch after having been reused repeatedly 5 times.
