老芒麦野生种质资源遗传多样性的SSR分析

摘    要:采用SSR标记技术,对来源于内蒙古5个盟市的7个样点的35份老芒麦(Elymus sibiricus)野生种质资源进行了遗传多样性分析.从筛选出多态性强、重复性好的20对引物中,检测出326个等位位点,平均每对引物产生12.55个多态位点;多态位点百分率为76.99%,遗传距离的变化范围在O.0000—0.3079之间,说明供试材料问亲缘关系较近;35份老芒麦种质的Nei’S遗传多样性指数(h)为0.2223,Shannon指数(I)为0.3288,意味着供试的35份材料间的遗传多样性相对丰富.在O.71水平处,测试材料大致分为两大类,来源于内蒙古东部、中部和西部区的材料交叉明显,表现出地理非同源性.本研究为探讨老芒麦遗传变异与环境的关系及老芒麦野生种质资源的保护提供了理论依据.

Abstract:Genetic diversity of 35 wild Elymus sibiricus germplasm resources from seven sample points of five union city in Inner Mongolia region was evaluated by using SSR markers.Twenty pairs of the SSR polymorphic primers were used in the SSR analysis. A total of 326 bands were detected, the average was 12.55 polymorphic bands per primer pair;the percentage of polymorphic was 76.99~ and the genetic dis- tance ranged from 0.0000 to 0.3079,indicating that the relationship amnong all individuals was close; The Nei~ s genetic diversity index (h) of 35 E. sibiricus germplasm was 0.2223,the Shannon index (I) was 0.3288. These results indicated that the genetic diversity among the tested materials are relatively abun- dant. The E.sibiricus could be classified into two groups at genetic similarity 0.71. The E.sibiricus materi- als collected from the eastern,central and western areas of Inner Mongolia showed cross-clustering charac- teristics,and they exhibited no relationship with their geographical distribution. This study has a theoreti- cal basis to explore the sibiricus genetic variation relationship with the environment and E. sibiricus wild germplasm resources protection.
