何首乌根和茎愈伤组织的诱导和二苯乙烯苷含量的检测

摘    要:以何首乌根和茎为外植体,通过正交试验筛选根和茎诱导的最佳培养基,结果表明在26℃和暗培养条件下,MS培养基中添加1 mg?L-1 2,4-二氯苯氧乙酸(2,4-D)和0.4 mg?L-1萘乙酸(NAA)最容易诱导出愈伤组织,诱导率可以达到96.7 %。通过HPLC测定了根和茎愈伤组织的二苯乙烯苷含量,分别为1.5mg/g和0.4mg/g。

Abstract:Root and stem of Polygonum multiflorum Thumb. as explants, orthogonal medium design are made to acquire the best callus induction medium. Results shows root and stem are most easily induced on MS medium containing 1 mg •L-1 2, 4-Dichlorophenoxyacetic acid (2,4-D) and 0.4 mg •L-1 NAA (NAA) at 26 ℃ and dark culture conditions, and the induction rate are 96.7 % under the condition. Stilbene glycoside content of root and stem callus were determined by HPLC, which are respectively 1.5mg/g and 0.4mg/g.
