牛松质骨脱细胞骨细胞外基质的制备

摘    要:为寻找适合骨移植的骨组织替代物.脱细胞骨细胞外基质(Acellular bone extracellular matrix,ABECM)制备最佳方法。方法:根据析因设计原理用不同浓度曲拉通X-100液分别与高渗和低渗液联合后对牛肱骨上端松质骨组织进行适当的脱细胞处理.制成脱细胞牛松质骨,再进行HE染色,Massion染色及扫描电镜观察,寻求最佳的曲拉通X-100的浓度,脱细胞时间及最佳的制备脱细胞牛松质骨的方法。结果表明:10%氯化钠与0.5%曲拉通X-100联合处理48小时的脱细胞牛松质骨未见细胞成分,细胞外基质基本结构未被破坏。由此可见本方法可以成功的制备ABECM,曲拉通X-100是制备ABECM良好试剂,ABECM是一种新型的理想的骨替代物,有广泛的应用前景。

Abstract:The paper is to find a new kind of bone substitute-acellular bone extracellular matrix(ABECM) available for bone implant and find the best methods of preparation of acellular bovince cancellous bone.Mathods:bovince upper humerus cancellous bone was approached acellularly by using 0.5%sodium chloride,10% sodium chloride and TritonX-100 according to the analytic factorial experiment.Specimens were stained with HE,Massion and observed with naked eyes,microscopy and scanning electron microscopy,to find the best mathod of ABECM.Results showed there were no cellular components in respectively histological slides of the bones,but the basic structure of extracellular matrix existed in 10% sodium chloride,0.5%TritonX-100 in 48hrs.These indicate that ABECM can be obtained successfully with this procedure,TritonX-100 is a good reagent for preparing ABECM.ABECM may be a new kind of ideal boned substitute and will be applied widely in the future.
