两种提取牛源大肠杆菌内毒素方法的比较研究

对提取牛源大肠杆菌内毒素粗制品的两种方法进行了比较,为获得含量较高的牛源大肠杆菌内毒素免疫原提供了简便方法和切实可靠的理论依据。实验采用热酚-水法和酚-氯仿-石油醚法提取牛源大肠杆菌内毒素,鲎试剂凝集活性和终点显色法测定热酚-水法提取内毒素粗制品含量约为0．16EU／ng。酚-氯仿-石油醚法提取内毒素粗制品含量约为0．31EU／ng。实验结果表明：酚-氯仿-石油醚法提取的内毒素含量比热酚-水法高,此法不需要透析袋透析,因此更适用于内毒素的大量提取。

Comparison of Two Methods for Extraction of Lipoplysaccharide of Escherichia Coli from Cattle

The paper aims at comparison of two methods for extraction of crude LPS of E.coli from cattle,in order to obtain a higher content of immunogenicity of LPS.The "hot phenol-water" method and the "phenol-chloroform-petroleum ether"method were used to extract the crude LPS of E.coli from cattle,the content of LPS was determined by the activity of agglutinating tachypleus amebocyte lysate(TAL) and the end point chromogenic TAL.The Results were that the content of LPS extracted by the "hot phenol-water" method and the "phenol-chloroform-petroleum ether"method were 0.16EU/ng and 0.31EU/ng.Conclusion The result showed that a higher content of LPS was obtained with the "phenol-chloroform-petroleum ether" method.This method does not need dialysis,so it's more convenient and more applicable to the massive extraction of LPS.
