辣椒疫霉菌蛋白质的聚丙烯酰胺凝胶电泳法鉴定

本文以辣椒疫霉菌(Phytophthora capsici)A~1和A~2两个交配型及寄生疫霉菌(P.micotianae Var.parasitica)A~1和A~2两个交配型为标准菌株,对从新疆石河子、塔城、吐鲁番、伊犁、哈密和昌吉六个不同地区辣椒病株上分离的疫霉菌株(分别编号为Ph—2、Ph—11、Ph—14、、Ph—15、Ph—16、Ph—17)的蛋白质,用聚丙烯酰胺凝胶电泳法进行了分析和比较。结果证明,这6个菌株的蛋白质电泳图谱与辣椒疫霉菌标准菌株一致,它们都有相同的5条重带和基本相同的3～4条弱带,而与寄生疫霉菌完全不同。说明该法可做为疫霉菌鉴定的重要辅助手段之一,实验中同一菌种不同交配型间的蛋白质电泳图谱完全相同,但6个菌株中Ph—2、Ph—14和Ph—16的弱带与标准菌株间稍有差异,是属于菌株间的差异还是其它原因有待进一步研究。

Identification of Pepper Phytophthora Blight by Protein PAGE

Polyacrylamide gel electrophoresis (PAGE) was employed to analyse and compare the protein of six isolates of phytophthora isolated from pepper diseased plant at six different areas in Xinjiang. The standard phytophthora were Phytophthora capsici Al and A2; Ph. nicotianae var. parasitica Al and A2. The results identified that the six isolate' s the same protein electrophoresis chart as standard Ph. capsici, they had five strong bands and three to four weak bands in common, but had different from Ph. nicotianae var. parasitica. It supported the use of this approach as an aid in identification of phytophthora. The protein electrophoresis chart of same species but different mating type were identical in experiment. The one weak band of Ph-2, Ph-14 and Ph-16 in six isolates was different from Ph. sapsici' s, this reason would be further studied.
