中华芦荟多酚氧化酶的分离纯化

摘    要:采用丙酮粉沉淀抽滤法从中华芦荟中提取多酚氧化酶.提取的粗酶液经30%~80%硫酸铵分级沉淀,透析除盐,DEAE-Sephadex A-50阴离子交换柱层析,后经PEG 6000浓缩,得到纯化的多酚氧化酶,其纯化倍数为13.28.纯化后的酶液经SDS-PAGE电泳分析,采用考马斯亮蓝R-250染色显示为单一蛋白带,分子量约为54.1 KD.

Abstract:Polyphenol oxidase(PPO)was extracted from Chinese Aloe with aoetone.The crude enzyme extract was fractionated with solid ammonium sulfate of 30%~90% saturation.After dialysis,the enzyme solution was purified by DEAE-Sephadex A-50 column chromatography.The enzyme was concentrated by PEG 6000.A13.28 fold purification of PPO was obtained.The result from polyacrylamide gelelectrophoresis of purified enzyme indicated that only one band was found using coomassie brilliant blue R250 stain.The molecular weight is about 54.1 KD.
