摘    要:基于DNA与对甲氧基苯甲醛的荧光猝灭效应,以甲氧基苯甲醛作为荧光探针建立了一种简便快速测定DNA的荧光分析方法.考察了pH、荧光探针浓度、温度、放置时间等条件对荧光探针及DNA测定的影响.该方法测定DNA含量的线性范围为51~1 000 ng/mL,检出限为30 ng/mL,10次测量500 ng/mL DNA的相对标准偏差为2.0%,可用于合成样品的测定.

Abstract:A symmetric emission spectrum and a broad,continous excitation spectrum was given by p-methoxybenzaldhyde.A rapid determination of DNA has been developed based on its fluorescence quenching ability on the endogenous fluorescence of p-methoxybenzaldhyde(I).The influence of pH,concentration of I,temperature and placing time on the determination were studied in detail.Under optimum conditions,the calibration curve is linear over the range 51.0~1000.0 ng/mL,with a detection limit of 30.0 ng/mL.The relative standard deviation of 500.0 ng/mL DNA is 2.0%.This method is simple,rapid and sensitive and shows promise in the application of synthetic samples.
