人工饲料育家蚕/杆状病毒表达猪瘟病毒囊膜糖蛋白E2基因

摘    要:以重组病毒Bm-BacPAK6-E2接种全龄人工饲料育及5龄桑叶育家蚕皓月×菁松幼虫、蛹,观察重组病毒的感染发病情况,调查病蚕、蛹的采血量并进行SDS-PAGE分析.结果表明,全龄人工饲料育家蚕接种重组病毒的成功率、采血量等指标均与5龄桑叶育相近,SDS-PAGE蛋白质电泳在家蚕幼虫、蛹血淋巴中均检测到1条与目的蛋白理论分子量(63 kD)相符的特异性蛋白条带,初步认为是重组病毒Bm-BacPAK6-E2的融合表达产物.

Abstract:Both F1 hybrid silkworm(Hao Yue X Jjng Song)fed with the fully artificial diet and the fifth instar with mulberry leaves were infected respectively with recombinant virus Bm-BacPAK-E2. After having observed the procedure of the diseased, and investigated the blood of diseased silkworm as well as comparatively analysis the SDS-PAGE electrophoresis of the blood samples, we found that the index of silkworm rearing with fully artificial diet,such as the infection rate of recombinant virus and blood sampling's quantity of protein expressed,is similar as the index of the fifh instar with mulberry diet, which can give much experience to manufacture through silkwom s baculovirus expression system. And we also found the obviously special band at about 63 kD in SDS-PAGE is accordant to the protein molecular weight deduced by target gene. That results shows the target gene has been expressed successfully and also the protein produced as we expected.
