基于特征图谱和多指标成分含量测定的不同产地天葵子质量分析

摘    要:采用HPLC建立天葵子药材特征图谱并同时测定其紫草氰苷与格列风内酯的含量。色谱条件为色谱柱TSKgel ODS-100V(250 mm×4.6 mm,5 μm),流动相乙腈-0.2%磷酸溶液,流速0.4 mL·min-1,检测波长258 nm。运用聚类分析法比较不同产地、不同批次间天葵子的药材质量,应用中药指纹图谱相似度评价系统确定8个共有峰,指认2个色谱峰,15批药材相似度都在0.90以上。本研究能够建立天葵子药材的特征图谱并进行含量测定,方法简便稳定,数据真实可靠,可以作为天葵子药材品质鉴别、质量控制及资源开发利用的参考依据。

Abstract:High performance liquid chromatography (HPLC) was used to establish the characteristic fingerprint of Semiaquilegiae Radix and to determine the content of lithospermoside and griffonilide. The chromatographic conditions were as follows: TSKgel ODS-100V (250 mm×4.6 mm, 5 μm), acetonitrile-0.2% phosphoric acid solution as mobile phase, flow rate of 0.4 mL·min-1, and detection wavelength of 258 nm. Cluster analysis was performed to compare the drugs of Semiaquilegiae Radix of different origins and batches. Eight common peaks were determined and two chromatographic peaks were identified with the use of the similarity evaluation system of traditional Chinese medicine fingerprint. The similarity among 15 batches of medicinal materials was more than 0.90. The method is simple and stable, producing authentic and reliable data. It can be used as the research evidence of quality identification, quality control, and resource development and utilization in Semiaquilegiae Radix.
