摘    要:比较蜣螂不同浓度醇提物抗良性前列腺增生的作用,并探讨相关机制,为蜣螂的开发利用奠定基础。采用皮下注射丙酸睾酮制造大鼠良性前列腺增生模型,以前列腺湿重、前列腺指数、血清酸性磷酸酶质量浓度(ρPAP)、雌二醇质量浓度(ρE2)、双氢睾酮浓度(DHT)(cDHT)、睾酮质量浓度(ρT)、表皮细胞生长因子(EGF)、转化生长因子-β1(TGF-β1)、碱性成纤维细胞生长因子(bFGF)及胰岛素样生长因子-1(IGF-1)为指标,考察蜣螂醇提物对良性前列腺增生的影响。结果表明良性前列腺模型造模成功,与模型组相比,85%乙醇提取物组能明显降低大鼠前列腺湿重与前列腺指数,明显降低ρPAP,ρE2,cDHT及ρT水平,明显降低EGF,bFGF及IGF-1的表达,提高TGF-β1的表达,可显著降低前列腺体密度,显著增加比表面及比膜面值。蜣螂85%乙醇提物具有良好的抗良性前列腺增生的作用,其机制与调节EGF,TGF-β1,bFGF及IGF-1的表达有关。

Abstract:The present study aimed to compare the effect and mechanism of different concentrations of ethanol extract obtained from Catharsisus molossus Linnaeus on anti-benign prostatic hyperplasia and to consequently provide insights for the further development and use of C. molossus L. A benign prostatic hyperplasia model was established in rats by the subcutaneous injection of testosterone propionate. We considered prostate wet mass,prostate index,the concentration of prostatic acid phosphatase (ρPAP), estradiol(ρE2), dihydrotestosterone (cDHT), and testosterone (ρT),and the expression of epidermal growth factor (EGF), transforming growth factor β1 (TGF-β1), basic fibroblast growth factor (bFGF), and insulin-like growth factor 1 (IGF-1) as indicators and evaluated the effect of ethanol extract on benign prostatic hyperplasia. The benign prostatic hyperplasia rat model was successfully constructed. Compared with the model group, 85% ethanol extract group exhibited significant reduction in the prostate wet mass and prostate index of rats as well as in the levels of ρPAPE2, cDHT, and ρT. Moreover, the 85% ethanol extract group showed a significant decrease in the expression of EGF, bFGF, and IGF-1 and an increase in the expression of TGF-β1. In the 85% ethanol extract group, a significant decrease in the density and significant increase in the specific surface area and specific membrane area of the prostate gland were observed. The 85% ethanol extract obtained from C. molossus L. demonstrated a good anti-benign prostatic hyperplasia activity, and its mechanism was associated with the regulation of the expression of EGF, TGF-β1, bFGF, and IGF-1.
