| 摘 要： | 以大连渤海湾分离260余株低温条件下生长良好的菌株为出发菌株,利用双层平板筛选方法选出2株低温蛋白酶产生菌(Bacillus subtilis).经UV、DES、NTG、EMS、LiCl单独及复合诱变,选育出一株(BS070623)蛋白酶高产突变株.通过单因素实验,确定了BS070623菌株蛋白酶发酵培养基为:玉米淀粉糖0.8%,豆饼粉1.4%,磷酸氢二钾1.0%,磷酸二氢钾0.7%.上述条件下该突变株低温蛋白酶产量为913.2 U/mg.

| Abstract: | Two strains are selected from the 260 low-temperature bacterial strains grown in Dalian's Bohai Sea bay,and thus by certain means,bacillus subtilis is bred.By single and multiple mutagenesis,such as UV,DES,NTG,MES,and LiCl,one mutant stain BS 070623 is selectively bred. By orthogonal experiment,the fermentation medium of BS 070623 is composed of 0.8%of corn syrup,1.4%of soymeal,1.0%of K_2HPO_4,and 0.7%of KH_2PO_(4-) Under the above condition,the production of the low-temperature protease is 913.2 U/mg. |
