芥菜原生质体培养和胚性愈伤组织的产生

摘    要:以芥莱无菌苗下胚轴为材料游离获得原生质体,原生质体培养于含NAA1.5mg/l,6-BA0.6mg/l、和2.4-D0.5mg/l的DPD液体培养基中,约4—5周形成肉眼可见的小愈伤组织。转入MS固体培养基后,即可产生大量的愈伤组织,其中胚性愈伤组织约占50%以上。

Abstract:Hypocotyl protoplasts from Brassica juncea (L.) Czern. et Coss were cultured in DPD liquid medium containing NAA 1.5rag/1, 6-BA 0.6rag/1, and 2.4-D 0.5mg/1.Protoplasts form microcolonies in 4-5 weeks and develop into a number of calli after being transfered to MS solid medium. About fifty percent of total calli are embryogenetic calli.
