摘    要:利用RAPD技术对普通烟草(Nicotiana tabacum L.cv Xanthi nc)和毛曼陀罗(Datura innoxia Mill)原生质体融合获得的体细胞杂种进行了分析.使用了9个10-mer随机引物对7个体细胞杂种和双亲进行RAPD指纹图谱分析,从7个杂种的9组DNA指纹图谱上共得到570条扩增带,其中与烟草特征谱带相同的带数占了67.8%,与双亲共有的谱带相同的带数为24.6%,而与曼陀罗特征谱带相同的带数仅有2.8%.由此可见,杂种组织倾向于排除毛曼陀罗的DNA,融合体是一个高度不对称的体细胞杂种.此外,杂种中还出现了双亲没有的谱带(新带占4.8%).

Abstract:The somatic hybrids between tobacco(Nicotiana tabacum L.cv Xanthi nc)and hairy datura(Datura innoxia Mill.)were obtained by PEG/DMSO fusion method.The somatic hybrids were identified with RAPD.Among a total of 570 bands observed from 7 somatic hybrid plants after DNAamplification with 9 arbitrary 10-mer primers,386(67.8 %) were tobacco-specific bands and 141(24.6 %)showed common bends of both the parents,only 16(2.8 %) homologue to hairy datura-specific bands.The results suggested that besides containing nuclear substances of two parents,the hybrid genomes were inclined to eliminate hairy datura chromosomes with DNA reconstruction.The hybrids were asymmetric.There were 4.8 % new bands(not seen in two parents)shown in the RAPD fingerprints of somatic hybrids.
