摘    要:以激光-紫外线复合诱变筛选的桑黄菌菌株S1为研究对象,通过单因素试验初步研究了液体发酵条件,根据二次回归旋转正交组合设计确定的最佳发酵条件为:装瓶量120 mL、接种量17 mL、发酵温度26℃、摇床转速135 r·min-1,并通过验证试验得到实际菌丝产量为(24.47 ±0.80)g·L-1.在此基础上进行了小型发酵罐试验,得到的最佳发酵条件为:搅拌速度180 r·min-1、温度29℃、空气流量6 L·min-1,得到的菌丝和胞外多糖产量分别为(68.23±2.51)g和(10.23 ±0.35)g.

Abstract:Phellinus igniarius strain S1 mutagenized with laser and UV was chosen to study the conditions of liquid fermentation by single-factor experiment.Based on quadratic orthogonal rotation combination design,the optimum conditions were determined as media amount of 120 mL with inoculum size of 17 mL,and fermentation temperature of 26 ℃ at rotate speed of 135 r·min-1.According to verification experiment,the actual mycelial yield was at the range of(24.47±0.80) g·L-1.The compact fermentor experiment was conducted based on the designed optimum conditions.The results show that the optimum fermentation conditions were with stirring speed of 180 r·min-1 at temperature of 29 ℃ with air flow of 6 L·min-1.The mycelia and expolysaccharide yields can reach the range of(68.23±2.51)g and(10.23±0.35)g,respectively.
