福建三尖杉中生物碱的高效液相分离与鉴定

摘    要:分别用半制备型、分析型反相高效液相色谱法(HPLC)对福建三尖杉中的生物碱进行分离和检测,并用液质联用(LC-MS)、核磁共振(NMR)等对纯化样品进行结构分析.实验得到了4种含量较高的三尖杉生物碱,并检测到了其他微量生物碱4个.其中,碱(m/z)344和碱(m/z)330与具有相同相对分子质量的已知生物碱的^1H NMR数据差异较大.与传统方法相比,反相HPLC分离检测方法简单高效,且对环境友好,对寻找新的微量化合物、提高资源利用率有重要意义.

Abstract:Reverse-phase semipreparative HPLC was used to separate alkaloids from Cephalotaxus fortunei. A HPLC method was established to analyze the products. Eight alkaloids were detected and four majors were isolated. Their structures were identified by LC-MS,~1H NMR and~(13)C NMR spectroscopic studies. Two alkaloids, having the same mol.wts as some known alkaloids', both showed different~1H NMR spectrum data. Comparing to traditional ones, the reverse-phase semipreparative HPLC method with a simple and high efficient flow is better and environmental benign. It will help to search for novel minor compounds and make use of limited plant resources with a greater efficiency.
