NGAL基因在原发性肝癌中的表达及其功能分析

摘    要:NGAL基因是Lipocalin家族成员,与MMP9的功能密切相关,在乳腺癌、食管癌等多种恶性肿瘤中异常表达,为了研究其在原发性肝癌中的表达及功能,通过荧光实时定量PCR法检测了NGAL基因在23对肝癌组织标本中的表达情况.结果显示:在12例肝癌组织中NGAL显著上调表达,占总样本数的52.17%(P〈0.05),提示了NGAL基因的表达水平可能与肝癌的发生发展过程有关.通过构建绿色荧光蛋白融合表达载体,用脂质体法转化肝癌细胞株SMMC-7721进行瞬时表达.细胞周期实验显示NGAL的表达可促使SMCC-7721细胞从G1期向S期转化,转化NGAL的细胞中S期细胞显著增多(P〈0.05),提示其可能具有促进肝癌细胞生长的功能;细胞凋亡实验发现NGAL的表达可显著抑制低血清诱导下的细胞凋亡率(P〈0.05),提示其可能参与抑制肝癌细胞的凋亡.

Abstract:NGAL(neutrophil gelatinase associated lipocalin) gene is a new member of Lipocalin family,and is closely related to the function of MMP-9.NGAL is expressed abnormally in several carcinomas,such as breast cancer and esophageal carinomas.To understand the expression and function of NGAL gene in Hepatocellular carcinomas(HCC),the expression level of NGAL in 23 pairs of HCC tissues was measured via Real-time RT-PCR method.The results indicated that NGAL was highly expressed in 12 HCC tissues,accounting for 52.17% of the total samples.Cell cycle experiment showed that overexpression of NGAL in SMCC-7721 is able to promote the cell from G1 to S phase,and the S phase is significant increased(P<0.05),which suggested that NGAL may play a role in hepatoma carcinoma cell growth.Also,Apoptosis experiment showed that overexpression of NGAL had the anti-apoptosis effect in SMCC-7721 under serum starvation(P<0.05).
