葛花对大鼠酒精性肝损伤的预防作用研究

摘    要:通过每日予大鼠白酒灌胃方法复制大鼠酒精性肝损伤模型,观察中药葛花对大鼠酒精性肝损伤的预防作用。110只雄性Wistar大鼠随机分为6组:空白组、模型组、葛花醇提物低剂量组、葛花醇提物中剂量组、葛花醇提物高剂量组和阳性对照药东宝甘泰片组。在造模的同时灌胃葛花醇提物及东宝甘泰片水溶液。连续灌胃8周,8周末取材观察:血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)活性,总蛋白(TP)及白蛋白(ALB)含量;肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、还原型谷胱甘肽(GSH)含量;肝组织病理变化。结果显示,模型组大鼠出现肝组织脂肪变性和炎性浸润,血清ALT、AST、ALP活性较空白组显著增加,TP、ALB含量较空白组显著降低;且肝组织中SOD活性显著降低,MDA含量显著升高。葛花醇提物组大鼠的肝组织脂肪变性和炎性浸润均较模型组有所减轻,血清ALT、AST活性较模型组显著降低,TP、ALB含量较模型组显著升高,肝组织SOD活性较模型组显著升高,MDA含量显著低于模型组,GSH含量较模型组也明显升高。研究表明,葛花醇提物具有明显预防酒精性肝损伤的作用。

Abstract:The alcoholic liver disease model rats were treated by feeding alcohol. To study the preventive effects of Flos Puerariae Lobatae on alcoholic liver disease of rats, 110 Wistar rats were randomly divided into 6 groups: normal group, model group, low, middle, high dosage of Flos Puerariae Lobatae groups and Compound Methionine and Choline Bitartrate Tablets group. Administration began at the same time of feeding alcohol for model group. Samples were taken at the end of 8th week. The followings were detected: the activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP); the contents of total protein (TP) and ALBumin (ALB); the activities of liver superoxide dismutase (SOD); the contents of liver maleic dialdehyde (MDA) and (GSH); and the liver pathology. Results in the experiment show that there were serious hepatic steatosis and inflammation in the model group. Activities of serum ALT, AST and ALP of the model group were significantly increased compared with the normal group. The contents of TP and ALB were significantly decreased compared with the normal group. There were significant decrease in SOD and significant increase in MDA. Ethyl alcohol extraction of Flos Puerariae Lobatae can obviously alleviate hepatic steatosis and inflammation in the model group. Ethyl alcohol extraction of Flos Puerariae Lobatae can significantly decrease serum ALT, AST activity, liver MDA content and increase the contents of TP and ALB. And it can also increase liver SOD activity, the content of GSH. Flos Puerariae Lobatae has good experimentally preventive effect on the Alcoholic Liver Disease.
