蓝隐藻藻蓝蛋白的分离、纯化及性质研究

摘    要:以实验室培养的蓝隐藻(Chroomonas placoidea)为材料,经过压力破碎和高速离心分离去除膜成分,得到的上清液分别采用50%、70%、80%、90%和100%的硫酸铵分级沉淀.沉淀经2次葡聚糖Sephadex G-100层析,得到了纯化的藻蓝蛋白,并对各级藻蓝蛋白的吸收光谱、荧光谱、亚基组成进行了分析.结果显示,隐藻藻胆蛋白呈现出很宽的硫铵沉淀范围,但各种沉淀样品具有几乎相同的光谱特性和相似的亚基组成,说明其不同的水溶性质可能与其亚基聚合程度不同有关;得到的藻蓝蛋白经HPLC和中性PAGE检测,基本不含杂质,A645/A280比值最高可达7以上.结果为今后的研究和应用奠定了基础.

Abstract:Chroomonas placoidea cells cultured in laboratory are broken and centrifuged to remove the membrane debris.The supernatants are precipitated by 50%,70%,80%,90% and 100% of ammonium sulfate respectively.Sediments containing PC are purified by two steps of Sephadex G-100gel filtration chromatography.The absorption spectra,fluorescence spectroscopy,as well as subunit composition of purified PC are determined.The results show that the PC of C.placoidea precipitates during a quite broad range of ammonium sulfate...
