苦瓜籽中一种抗胰蛋白肽的分离及抗真菌作用观察

摘    要:利用凝胶过滤层析Sephadex G-75、强阳离子交换色谱SP-650M,从苦瓜籽中分离纯化出一种活性多肽.该多肽在Tricine-SDS-PAGE上显示为单一的谱带,分子量约为6.8 kDa.在经还原剂二硫苏糖醇(DTT)处理后的电泳结果表明该多肽不含有分子间的二硫键.此多肽是一种胰蛋白酶抑制剂,同时具有抗植物致病菌活性,是一种具有双功能作用的多肽.

Abstract:A novel antifungal peptide was isolated from bitter melon seeds by extraction with PBS,Sephadex G75 chromatography and strong cation exchange chromatography column(SP-650M).It was identified to be homogeneous by Tricine-SDS-PAGE,and the relative molecular weight was estimated to be about 6.8kDa.It was a trypsin inhibitor and showed antifungal activity against Pythium aphanidermatum,Fusarium oxysporum and Botrytis cinerea.It is expected to be applied to the prevention of insect pests and plant diseases.
