庆大霉素发酵液中C组分含量测定

摘    要:本文用荧光薄层扫描法测定庆大霉素发酵液中C组分含量。用氯仿:甲醇:氨水=4:5:5下相 为展开剂,在硅胶G-CMC板上分离庆大霉素C1,C2及C1a,喷甘油作增荧光试剂,然后用薄层 扫描仪测定其荧光强度。本法具有快速、简便、灵敏度高(C1,C2及C1a的检出限量分别为 0.3、 0.7及0.2μg),线性范围宽及重现性好等优点。也可用于成品及半成品测定。

Abstract:A fluoro-densitometric method is reported for the quantitative determination of gentamicin C-compount in fermentation broth. The gentamicin C1. C2and C1a were separated on a silica gel CMC plate by using lower phase of chloroform- methanol - concentrated ammonium hydroxide(4:5: 5 ) as developer, and sprayed flu- orescence-cnhancing reagent: glycerol. The fluor-densitometry of C1, C2and C1a were determined with a thin layer chromatographic scanner. This method is quick, simple, high sensitivity (The measurable lowest measures of C1, C2, andC1. are 0. 3, 0. 7 and 0. 2 ug respectively ), and of wide range of linearity and good reproductibility.
