黄芪注射液对急性肺损伤大鼠肺组织中存活素表达的影响

摘    要:目的探讨黄芪注射液对急性肺损伤大鼠肺组织中存活素表达的影响。方法将36只sD大鼠按随机数字表分为3组,各组注药均从尾静脉注入。对照组注入生理盐水1ml;模型组应用脂多糖5mg/ks溶解于1ml生理盐水注入;干预组在模型组基础上应用黄芪注射液治疗。24h后处死,收集右上肺组织固定包埋,常规HE染色,观察肺组织病理变化。用免疫组化和图像分析系统定量检测肺组织中存活素表达。用TUNEL原位凋亡法检测肺组织细胞凋亡,并计算凋亡指数。结果干预组肺组织炎症改变明显轻于模型组。急性肺损伤大鼠肺组织存活素表达为对照组最高,其次为干预组,模型组最低;而肺组织细胞凋亡指数为模型组最高,其次为干预组,最低为对照组,差异均有统计学意义。存活素与凋亡指数呈负相关性。结论黄芪注射液可能通过调控ALI大鼠肺组织中存活素表达,从而抑制肺组织细胞凋亡,而且黄芪注射液可能有改善ALI肺组织的病理改变。

Regulation of astragalus membranous injection and its effects on Survivin in lung tissue of acute lung injury mice

Abstract:Objective To study the regulation of astragalus membranous injection and its effects on Survivin in lung tissue of acute lung injury ( ALl ) mice. Method 36 Sprague Dawley male mice were divided into three groups. Normal groups were injected 1 ml saline from the caudal vein of mice. Model groups were injected lipopolysaccharide (5 mg/kg) which dissolved in lml saline from caudal vein of mice. Interventive groups used astragalus membranous injection treatment on the foundation of model group' s. All mice were killed after 24h, collected right upper lung tissue fixation embedding HE staining to observe pathol immunohistochemistry and quntitive image analysis system Survivin expression in tected by TUNEL assay and calculated the apoptotic index. Results Interventive in lung tissue were significantly lighter than model groups. ogical change in lung tissue. By lung tissue. Apoptosis were de groups in inflammatory changing ALI in rat lung tissue expression of Survivin in the nor mal groups were higher than that in the intervention groups and model groups; and the apoptosis index in lung tis sue in model groups were higher than that in the intervention groups and normal groups, and the differences were statistically significant. The Survivin and apoptosis index were the reverse changing, and correlation analysis showed a negative correlation. Conclusion As of ALl to suppress lung tissue apoptotic, and mice. tragalus could upper regulate the expression of Survivin in ung tissue astragalus could improve pathological changes of lung tissue in ALI
