尼莫地平和银杏叶提取物对谷氨酸神经毒性的对抗作用:形态学观察

摘    要:用形态学的方法观察了尼莫地平和银杏叶提取物(含黄酮甙类24%和内酯3.4%)对谷氨酸神经毒性的影响。结果发现谷氨酸导致新生小鼠下丘脑弓状核变性、坏死,钙通道拮抗剂尼莫地平及血小板激活因子受体拮抗剂银杏叶提取物均可剂量依赖性地抑制谷氨酸引起的神经元损伤。结果提示钙离子超载及血小板激活因子的激活与谷氨酸的神经毒性作用有关

Abstract:Protective effects of nimodipine and extracts of ginkgo biloba (EGb) from glutamate neurotoxicity in mice were morphologically evaluated. Neonatal mice were daily injected ip with saline, monosodium glutamate (1.0g/kg,MSG), nimodipine (0.1,0.5,1.0,1.5,2.0mg/kg, Nim)+MSG, EGb(1.0,3.0,5.0,7.0,10.0mg/kg)+MSG for 7 days, respectively. Nim or EGb was injected 15 min prior to MSG. The results showed that MSG could get rise to neuronal necrosis in arcuate nucleus (AN) of hypothalamus. Treatment of Nim prior to MSG decreased the number of necrotic neurons in the AN induced by MSG. Nim provided a dose dependently protective effect on the AN. Pretreatment with EGb obviously attenuated neuronal swelling and degenerating. EGb had the same dose dependently protective effect on the AN as Nim. Both Nim and EGb had protective effects on the AN lesions caused by MSG.
