摘    要:探讨中华补血草水提物(LSE)对抗N-乙酰对氨基酚(N-acetyl-p-aminophenol,APAP)引起的肝细胞损伤的作用及其可能的线粒体机制.通过分组实验,测定血清中谷丙、谷草转氨酶(sALT,sAST)的活性和谷胱甘肽(GSH)的含量,同时制备和观察肝组织切片.其后进一步探讨线粒体在这一过程中所发生的变化,通过测定线粒体的肿胀、线粒体膜电位(MMP)的变化以及电压依赖性离子通道(VDAC)在转录水平的变化等,探讨其相应的线粒体作用机制.结果显示:100、200、400mg/kg的LSE可以剂量依赖性地对抗APAP引起的肝细胞损伤,而这种护肝活性可能与LSE对肝细胞线粒体的保护作用有关.同时还发现,线粒体的一个重要的外膜蛋白——VDAC的表达可能与之有着密切的关系.

Abstract:The present study was undertaken to investigate the hepatoprotective activity of aqueous extracts of Limonium sinense (Girard) Ktze root (LSE) against acetaminophen (APAP)-induced liver damage and to elucidate the possible mechanisms underlying the hepatoprotection.
