用ELISA法测定SPA与多种动物IgG的结合特性

应用酶联免疫吸附试验对兔、大鼠、小鼠、小牛、绵羊、鲫鱼、鸭、蟾蜍和黄鳝9种动物血清IgG与SPA的结合特性进行了研究。酶标SPA浓度梯度,IgG包被浓度梯度及特异性自体和异体IgG阻断试验的结果表明:兔、大鼠、小鼠、小牛、绵羊及鲫鱼的IgG能与A蛋白结合,而鸭、蟾蜍及黄鳝的IgG不能与A蛋白结合。这些结果对进一步用葡萄球菌A蛋白作免疫测定和纯化IgG具有实用意义。

BINDING PROPERTIES BETWEEN STAPHYLOCOCCAL PROTEIN A (SPA) AND SERUM IgG OF SOME ANIMALS SPECIES

A direct SPA-ELISA had been established to study the binding proper--ties between staphylococcal protein A (SPA) and serum IgG of rabbit, rat, mouse, calf, sheep, crucian, carp,duck, toad and ricefield eel. Experimental results showed that serum IgG of rabbit, rat, mouse, calf, sheep and crucian carp had the binding propefty with SPA, while the IgG of duck, toad and ricefield eel had not. Experimental results also showed that the O.D. value varied with different working dilutions of HRP-SPA as the coating IgG con-centration fixed and depending on the concentrations of coating IgG the O.D. value also changed as the HRP-SPA workine dilution fixed. The binding property could be significantly reduced if we first mixed the IgGs with HRP-SPA and incubated, the mixture overnight before being used for testing IgG-binding properties.
