用Asmus法测K_m值

摘    要:在酶的动力学研究中,K_m能看作为表观电离常数,它在生物化学中是用米氏方程测定,只要酶和底物结合生成一个有色中间络合物,则K_m能用Asmus法测定,本文在酪氨酸酶催化多巴的反应中,用Asmus法计算K_m值,其结果与米氏方程式相一致。

Abstract:In study of enzyme kinetics, according to the principle of steady state K_m can be regarded as the apparent dissociation constant and determined using Michaelis equation. When enzyme binds with the substrate forming a colored intermediate complex, the value of K_m can be determined by Asmus method. This paper deals with the calculation of K_m in tyrosinase-catalyzed reaction with DOPA by Asmus method. Results obtained agree with that by Michaelis equation.
