铁皮石斛不同生长阶段遗传稳定性的RAPD分析

摘    要:利用从20个随机引物筛选出来的3个稳定性较好的10碱基引物对已继代12到13次仍然维持在不同生长阶段、来自同一蒴果的铁皮石斛T31种群进行RAPD检测其遗传稳定性,发现不同生长阶段内遗传相似系数较高,在89.36%和100%之间,其中原球茎期的变异比萌芽期、一叶期、两叶期、开花期要显著,但程度都极其微弱.

Abstract:To research the hereditary stability of Dendrobium officinale Kimura et Migo which were subcultured 12 ~ 13 times in the same tissue culture system from five different development stages, 3 ten - base primers screened from 20 arbitrary primers were used for DNA amplification with random amplified polymorphic DNA (RAPD). The results showed that the genetic similarity coefficient are very high which varied from 89. 36% to 100%. The variation of protocorm were rather more remarkable than that of germination, monoleaf - stage, bileaf - stage and inflorescence.
