真菌α-淀粉酶高产菌株的诱变选育研究

摘    要:以米曲霉2197为出发菌株,采用紫外线、硫酸二乙酯6、0Co-γ射线、亚硝酸、微波及离子束交替进行的复合诱变育种技术,通过平板初筛、固体麸皮初筛、固体麸皮复筛,获得一株真菌-α淀粉酶高产菌株ZLF 13,并对其进行了生活力试验和遗传稳定性试验.结果表明,突变株ZLF 13生长代谢旺盛,营养要求低,遗传稳定性好,产酶水平较出发菌株提高6.4倍,达946 u/g.

Abstract:Mutant(ZLF 13)producing high-yield fungus α-amylase was obtained from aspergillus oryzae(2197)by multiple mutagenesis(UV,DES,60Co γ-rays,HNO2,micwaves and ion beam),isolating and screening.After experiment on life activity and genetic stability,the results indicated that mutant(ZLF 13) has terrific characters such as active metabolism,lower minimal needs of nutrition and genetic stability.The highest level of fungus α-amylase reached up to 6.4 times which compared with the original stains′.The level of producing can be utilized in industrial production of fungus α-amylase.
