棋盘法和倍比微量稀释法在体外联合抗真菌活性药物筛选中的应用研究

摘    要:通过对氟康唑和盐酸小檗碱体外联合抗白色念珠菌活性检测,比较棋盘法和倍比微量稀释法在体外联合抗真菌药物筛选研究中的应用,优选更适合实验室体外联合抗真菌药物通量筛选的方法.分别用2种药物筛选方法检测氟康唑和盐酸小檗碱联合对白色念珠菌的活性,计算抑菌联合指数,其抑菌联合指数(FICI)0.5,则表明两药具有协同作用.棋盘法和倍比微量稀释法均能验证氟康唑和盐酸小檗碱联用具有协同作用,但倍比微量稀释法操作更简单,抑菌联合指数的计算更直观,更适用于大批量药物的筛选.倍比微量稀释法更适合于实验室体外联合抗真菌药物的通量筛选.

Comparison of the checkerboard method and the multiple microdilution method based on drug screening in combined treatment of anti-Candida albicans

Abstract:To compare the application of the checkerboard method and the multiple microdilution method in antifungal drug screening, in-vitro anti-Candida albicans activity of fluconazole or berberine hydrochloride was tested and the better method for laboratory antifungal screening was selected. The activity of fluconazole and berberine hydrochloride against Candida albicans was evaluated by two drug screening methods respectively followed by calculation of the combined inhibition index(FICI). The FICIs of both drugs was less than 0.5 with synergistic effects. The checkerboard microdilution method and the multiple microdilution method can verify the synergistic effect of fluconazole and berberine hydrochloride, but the latter has a simpler operation procedure, and the calculation of FICI is more objective and suitable for large-scale drug screening. The multiple microdilution method is better for in-vitro screening of antifungal drugs.
