剑叶龙血树树叶化学成分及其改善HepG-2细胞胰岛素抵抗的作用

摘    要:采用硅胶、Sephadex LH-20和反相RP C18等柱色谱技术,从云南剑叶龙血树(Dracaena cochinchinensis)树叶甲醇和石油醚提取物中分离了7个化合物,包括3个黄酮类化合物,1个三萜和3个甾体化合物,经综合波谱分析分别鉴定为:4',7-二羟基-3'-甲氧基-8-甲基黄烷(1),7-羟基-4'-甲氧基高异黄烷(2),2',4,4'-三羟基查尔酮(3),(22E)-3β-acetoxystigmasta-5,22-diene(4),lanost-9-en-3β-ol(5),β-谷甾醇(6),胡萝卜苷(7).测定了化合物1~3改善Hep G-2细胞对胰岛素抵抗的作用,化合物1~3能显著增加细胞的葡萄糖消耗量.

Chemical constitents of leaves from Dracaena cochinchinensis and their ameliorative effects on insulin resistance in Hep G- 2 cell

Abstract:This research focused on the chemical constituents isolated from the leaves of Dracaena cochinchinensis( Lour.) S. C. Chen. Seven compounds were obtained from the methanolic and petroleum ether extracts by various column chromatography on Si gel,Sephadex LH- 20 and RP C- 18,respectively. Their structures were identified as4',7- dihydroxy- 3'- methoxy- 8- methylflavan( 1),7- hydroxy- 3-( 4'- methoxybenzyl)- 4- chromanone( 2),2',4,4'- trihydroxychalcone( 3),( 22E)- 3β- acetoxystigmasta- 5,22- diene( 4),lanost- 9- en- 3β-ol( 5),β- sitosterol( 6),daucosterol( 7),based on the combined spectroscopic data and by comparison of these data with values from literatures. Compounds 1—3 were tested for their ameliorative effects on the insulin resistance in Hep G- 2 cell. Compounds 1—3 displayed relatively strong activities.
