紫薇受粉习性及花粉管生长的研究

摘    要:用脱色苯胺蓝对紫薇花粉和花粉管进行染色,在荧光显微镜下观察其自然条件下的受粉习性及花粉管生长行为,以确定花粉管通道的形成时期.结果表明,盛花期的紫薇在开花后1.5h内完成受粉,花粉落量多,受粉率为100%;开花后24h左右进行受精;单果种子数为36~50粒,认为可以用花粉管通道法转化紫薇,导入外源DNA的最佳时机为开花后24h左右.

Abstract:Lagerstroemia indica L. was used as plant material. To study the pollinating habit in natural conditions and the course of pollen tube growth, the pollen grain on stigma and the pollen tube growth in style were monitored by means of fluorescence microscope after decolorized aniline blue staining. The special attention was paid to the determination of time when the pollen tube entered into the ovule. It was concluded that pollination, with a huge amount of pollen grains on stigma and 100% pollination rate, was completed within 1.5 h after flowering during its profuse flowering period, the fertilization was present about 24 h after flowering, the quantity of seed per fruit was 36-50. So Lagerstroemia indica L. could be the ideal plant material suitable for transformation by the technique of introducing exogenous DNA into the ovule via the pollen-tube pathway. The optimum time of introducing exogenous DNA into its ovule was approximately 24 h after flowering.
