红豆杉愈伤组织培养中褐变现象的初探

摘    要:多酚氧化酶(PPO)和过氧化物酶(PO)的性高低与红豆杉愈伤组织的褐变程度有着密切关系,即酶活性高,愈伤组织褐变严重,通过在培养基中添加适量的Vc、活性炭、CA、GA3和ABA等物质,能抑制酶的活性,减轻褐变程度,提高愈伤组织的生物产量。

Abstract:The activities of PPO, especially PO were relevant to the darkening in Taxus chinensis var. mairei tissue cultures. With the improvement in the activity of PPO and PO, the darkening degree became more and more serious and the growth of callus was inhabited. The activities of PPO and PO were decreased by adding optimum concentration of vitamin C, casein hydrolysate (CA), activated carbon and other factors such as GA 3, ABA, glucose & fructose to B 5 medium. And the degree of darkening would be mitigated abviously. The growth of callus was promoted accordingly.
