文殊兰弱碱性生物碱的抗肿瘤活性研究

摘    要:研究文殊兰中弱碱性生物碱对不同肿瘤细胞的抑制作用,并筛选出敏感细胞株,观察凋亡形态;MCF-7细胞株、SMMC7721细胞株、SGC7901细胞株、HepG-2细胞株,MTT溶液,DMSO,Hoechst33258等;利用MTT法检测文殊兰弱碱性生物碱对上述肿瘤细胞的抑制作用以及Hoechst 33258染色法观察细胞凋亡的形态学变化;文殊兰弱碱性生物碱对上述细胞株均有明显的抑制作用,对HepG-2细胞的抑制作用更为明显,给药72 h后测定IC50=9.374 323μg/mL;Hoechst 33258染色法观察细胞出现凋亡小体,有凋亡现象发生.文殊兰弱碱性生物碱MCF-7细胞株、SMMC7721细胞株、SGC7901细胞株、HepG-2细胞株具有不同程度的抑制作用,且呈现一定的剂量依赖性,对HepG-2细胞的抑制作用最强,表现出良好的抗肿瘤活性.

Abstract:Research the anti-tumor activity of Alkalescent alkaloids from Crinum .And screen the sensitive cells then observe morphological changes of cell apoptosis; MCF -7 cell, SMMC7721 cell, SGC7901 cell, HepG-2 cell, MTT solution, DMSO,Hoechst 33258;ex-periments could illustrate the use of the drug inhibition of these tumor cells by MTT assay .It could observe morphological changes of cell apoptosis by Hoechst 33258 staining; Alkales-cent alkaloids from crinum could inhibit these cells obviously .HepG-2 cell was inhibited more sensitive to alkaloids .After dosing 72 h it showed that IC 50 =9.374323μg/mL;it can be seen apoptotic bodies and apoptotic phenomenon by Hoechst 33258 staining;the results indicated that the Alkalescent alkaloids from crinum could inhibit the MCF -7, SMMC7721, SGC7901 , HepG-2 cells significantly .The suppression was in a dose dependent manne . The inhibition of HepG-2 was the strongest .It could show the excellent anti-tumor activity.
