大孔树脂纯化八珍汤苷类部位的研究

摘    要:采用大孔吸附树脂技术对苷类部位进行纯化.以芍药苷、人参皂苷Rb1及总苷(以芍药苷计)、总皂苷(以人参皂苷Re计)的含量为指标,筛选了8种大孔树脂,优选出吸附容量及洗脱效果较好的D-101型大孔树脂,通过对吸附动力学曲线、药液pH值、漏点质量浓度的考察,确定最佳吸附条件为药液质量浓度0.25 g/mL,药液pH值为6,吸附时间为8 h;通过对洗脱液质量分数、洗脱液pH值、洗脱液体积、洗脱液流速的考察,确定最佳洗脱条件为洗脱液质量分数为80%的乙醇,pH值为8,洗脱液体积为40 BV,洗脱流速为3 BV/min.芍药苷、人参皂苷Rb1及总苷(以芍药苷计)、总皂苷(以人参皂苷Re计)的洗脱率分别为98.14%、96.83%、91.71%、91.21%,终产品中总苷类成分的百分含量占总洗脱物的24.44%、总皂苷类成分的百分含量占总洗脱物的68.90%.
