功劳木有效成分逆转肿瘤多药耐药研究

摘    要:采用高内涵活细胞成像系统检测功劳木10种有效成分对肿瘤细胞K562/ADM细胞内罗丹明(rhodamine123,Rh123)蓄积的影响,结果发现化合物2-9可以不同程度地提高K562/ADM细胞中Rh123的荧光强度(给药浓度为10μmol/L时,Rh123荧光强度增长率分别为15.19%、24.01%、8.70%、73.17%、21.84%、76.98%、17.26%和23.29%).结果表明化合物2-9均具有逆转肿瘤多药耐药活性,其中化合物7的抗多药耐药活性最强.

Abstract:To research reversal of multidrug resistance by 10 compounds from the stem of Mahonia bealei(Fort.) Carr.,the effects of 10 compounds on rhodamine123 accumulation in K562/ADM cells were detected by high content screening analysis.Compound 2-9 can raise mean fluorescence intensity(MFI) of rhodamine123 in K562/ADM cells by different degrees(MFI increasing rate of Rh123 were 15.19 %,24.01 %,8.70%,73.17 %,21.84 %,76.98 %,17.26 % and 23.29 % at a concentration of 10 μmol/L respectively).Compound 2-9 possess the reversal of multidrug resistance activity,among which compound 7 was the strongest.
