重组人白细胞介素6工程菌的表达和功能初探

自人外周血的ｍＲＮＡ中获得了白细胞介素６ｃＤＮＡ，基国被克隆并在大肠杆菌中表达，在２０Ｌ发酵罐中每Ｌ发酵液可得６０ｇ湿菌体，ＩＬ－６的表达量约占大肠杆菌全蛋白的３０％以上，占包含体蛋白的６０％。纯化产物在长期，急性毒性和三致实验正常的基础上，用恒河猴的功能实验表明，在不制造血象降低模型的情况下，仍能明显增加血液中血小板和白细胞的数量。

Expression of Engineering Bacterium and Function of Recombinant Human Interleukin-6(IL-6)

Interleukin\|6(IL\|6) cDNA was obtained from the mRNA of human peripheral blood. Gene was cloned and expressed in \%E.coli.\% Sixty grams wet pellet was obtained from per liter fermentation liquid in 20L fermentation equipment. There were more than 30% IL\|6 protein in the whole protein of the bacterium and 60% IL\|6 protein in the inclusion body. Upon the basis of normal of long toxicity and short toxicity, the experiment on rhesus monkey shows that IL\|6 can increase the quantity of platelet and leukocyte obviously without the model of decreasing hemogram.
